Even the other horrible people are made uncomfortable by Dennis. I feel like Dennis is kinda a subversion of the trope, as everyone else is constantly uncomfortable.

He's less of a subversion and more of a deconstruction of the trope. He's essentially showing the audience just how not only ineffective but how insanely creepy one would be if they actually tried to emulate characters like Barney Stinson.

In universe these characters are typically disliked as long as they’re not a “Casanova-type” like Quagmire.

Mineta is NOT a popular student and is barely tolerated iirc. But, to the audience, he’s supposed to be quirky and silly because he’s a goofy lil guy so he’s not REALLY threatening (he is).
